Sanger Sequencing and Fluorescence Spectroscopy

Sanger sequencing is a DNA sequencing technique using the principle of Fluorescence spectroscopy. After the amplification step, PCR is halted by the addition of ddNTPs tagged with different fluorophores. The DNA fragments are arranged in size by capillary gel electrophoresis, passing in front a laser excitation. Specific fluorescence bands are emitted and the resulting chromatogram is processed for analysis.
